PHARMACOLOGICAL ACTION

GLUCOSAMINE: Glucosamine is a derivative of cellar glucose metabolism and is a dietary supplement not a pharmaceutical drug. It is also a component of glycosaminoglycans and proteoglycans of the cartilage matrix covering the ends of bones and hyaluronic acid which is a part of synovial fluid within the joint. The mechanism of action of glucosamine in human is unknown. Because glucosamine is a part of the cartilage matrix in joint tissues, it has been theorized for many years that its administration could effect symptomatic relief for Osteoarthritis sufferers by supplying the components for cartilage repair and thus reduce pain and disability.

CHONDROITIN SULPHATE: The 3 proposed mechanisms of action of chondroitin sulphate are (1) anti-inflammatory activity, (2) effecting the synthesis of hyaluronic acid, and (3) “the direct antidegradative actions” of chondroitin sulfate.

FISH OIL OMEGA 3: Fish oil most potent effect on artherosclerosis may be related to its potential to alter plaque inflammation, thereby stabilizing vulnerable plaques. In recent years there has been a growing body of evidence that is shifting the paradigm of how inflammation is contained and dissipated. In this new model, inflammation resolution is an active process mediated by lipid-derived compounds. Newly discovered families of chemical mediators, resolvins and protectins are directly involved in blocking neutriphil migration, infiltration, and recruitment, as well as in blocking T-cell migration and promoting T-cell apoptosis. In addition, protectins can reduce tumor necrosis factor and interferon secretion.

PHARMACOKINETICS

GLUCOSAMINE

Orally administered glucosamine is rapidly absorbed, highly distributed and efficiently cleared. The gut rather than liver is mainly responsible for the first pass metabolism since reduced bioavailability is observed after oral but not intraperitoneal doses.

CHONDROITIN SULPHATE

The extent of absorption of chondroitin sulfate as indicated by the mean Cmax (21.5 µg/ml) and mean AUC (187 µg/ml h) of total disaccharides after dosing (1600 mg) provides evidence that chondroitin sulfate is absorbed orally. The bioavailability of CS ranged from 4.8 to 5.0% after single dosing and 200-278% upon multiple dosing. Chondroitin sulfate is bioavailable after oral dosing.
In addition, the low molecular weight chondroitin sulfate used in this study displays significant accumulation upon multiple dosing.

INDICATIONS

Formulated for symptoms associated with aging, injury of the bones, joints, muscles and tendons. Also indicated in problems like osteoarthritis, knee pain and back pain.

DOSAGE AND ADMINISTRATIONS

Take up to two capsules daily prior to meal. Do not exceed the recommended dosage. Food supplements are intended to supplement the diet and should not be regarded as a substitute for a varied diet and healthy lifestyle. •Patients taking blood-thinning medications (anticoagulants) or daily aspirin therapy.

PRECAUTION AND WARNINGS

•Children, as well as women who are pregnant and/or nursing, or who become pregnant.

•Patients who have any adverse reactions or unusual side effects to glucosamine and/or chondroitin sulphate and/or Omega 3 supplements, should discontinue use immediately and consult their physician.

•The administration in patients with severe hepatic or renal insufficiency should be made under medical supervision.

•Derived from Seafood therefore should not be given to patients who are allergic to shellfish.

•A doctor should be consulted in order to exclude the presence of other joint conditions/diseases or which an alternative treatment should be considered.

PREGNANCY AND LACTATION

Available evidence is inconclusive or inadequate for use in pregnant or lactating mothers. Administration during the first 3 months of pregnancy must be avoided.

DRUG INTERACTIONS

Warfarin (Coumadin) interacts with Glucosamine, Chondroitin Sulphate and Omega 3 combination. Warfarin (Coumadin) is used to slow blood clotting. There are several reports showing that taking glucosamine with or without chondroitin increase the effect of warfarin (Coumadin) on blood clotting. Medications for cancer (Antimitotic chemotherapy) interacts with Glucosamine, Chondroitin Sulphate and Omega 3 combination. Acetaminophen (Tylenol, others) interacts with Glucosamine, Chondroitin Sulphate and Omega 3 combination. Medications for diabetes (Antidiabetes drugs) interacts with Glucosamine, Chondroitin Sulphate and Omega 3 combination. Acarbose, Acetohexamide, Chlorpropamide, Glipizide, Glyburide, like antidiabetics may also show some interactions. But more information is needed to know if this interaction is a big concern.

ADVERSE EFFECTS

Get emergency medical help if you have any of these signs of an allergic reaction while taking chrodroitin sulphate/glucosamine/Omega 3: hives;  difficulty breathing; swelling of your face, lips, tongue, or throat.

Less serious side effects may include: mild nausea, upset stomach; heartburn; or diarrhoea. Taking glucosamine and/or chondroitin sulphate and/or Omega 3 with food seems to reduce the incidence of the above side effects. To date, no significant side effects have been reported; although long-term studies on the safety of glucosamine, chondroitin sulphate and Omega 3 supplements still need to be completed.

OVER DOSAGE/TREATMENT

No cases of accidental or intentional overdose are known: The animal acute and chronic toxicological studies indicate that toxic effects and symptoms of toxicity are not likely to occur, even after high overdoses.
